RA Test - Rheumatoid Arthritis test यदि  जोड़ों के हड्डियों  में  सूजन लगती है, और  सुबह में विशेष रूप से कठोर हो जाते हैं,  संदेह हो सकता है कि आपको रुमेटीइड गठिया (आरए) है।यदि समय में इसका निदान और उपचार नहीं करते हैं, तो यह जोड़ों को नुकसान पहुंचा सकता है rheumatoid arthritis के  रोगियों के blood में  rheumatoid  कारक एक antibody मौजूद होते है। blood test  द्वारा   rheumatoid factor  के स्तर को मापते हैं।  positive rheumatoid factor test  का अर्थ है कि उनके blood में रोगी के संधिशोथ का स्तर उच्च माना जाता है। positive rheumatoid factor test  का उपयोग डॉक्टरों को निदान तक पहुंचने में सहायता करने के लिए एक सहायक  के रूप में किया जाता है।

Qualitative Method

1. Kit के साथ दिए गए  disposable slide पर marker pen  से,  T ( Test ) , PC ( Positive control ) और  NC (  Negative control ) लिखे |

2. Kit के साथ दिए गए  disposable pipette का उपयोग करके slide के  T ( Test) पर  test serum की एक बूंद डालें |

3. इसी तरह  slide के  PC ( Positive control ) पर kit के साथ दिए गए Positive control की एक बूंद डालें | और  NC ( Negative control ) पर kit के साथ दिए गए Negative control की एक बूंद डालें |

4. latex reagent की एक बूंद को serum के ऊपर,एक बूंद Positive control के ऊपर,एक बूंद Negative control के ऊपर डाले । dropper के   tip को तरल पर  touch होने  न दें।

5. Mixing stick का उपयोग करके, serum and latex reagent  को पूरे circle पर मिलाएं। इसी तरह PC और NC को भी mix कर लें |

6. तुरंत एक stopwatch प्रारंभ करें धीरे-धीरे slide को आगे पीछे  rotate करे , दो मिनट में  agglutination  के लिए देखें ।

Interpretation Of Result

Agglutination is a positive test result and indicates presence of rheumatoid factors in the specimen.

 No agglutination is a negative test result and indicates absence of rheumatoid factors in the specimen

Semi Quantitative Method

 सबसे पहले qualitative method से positive serum sample का  isotonic saline से  serial dilution  -    1:2,  1:4,  1:8,  1:16, 1:32,  1:64  बना ले |

1. Kit के साथ दिए गए  disposable slide पर marker pen  से,  1:2,  1:4,  1:8,  1:16, 1:32,  1:64 लिखे |

2. Kit के साथ दिए गए  disposable pipette का उपयोग करके slide के  1:2,  1:4,  1:8,  1:16, 1:32,  1:64  पर diluted test serum की एक एक drop डालें |

4. latex reagent की एक एक  drop को serum के ऊपर डाले । dropper के   tip को तरल पर  touch होने  न दें।

5. Mixing stick का उपयोग करके, serum and latex reagent  को पूरे circle पर मिलाएं। 

6. तुरंत एक stopwatch प्रारंभ करें धीरे-धीरे slide को आगे पीछे  rotate करे , दो मिनट में  agglutination  के लिए देखें ।

Interpretation Of Result

Agglutinations in the  highest serum dilution corresponds to the approximate amount of rheumatoid factors ( RF ) in IU/ml present in the test specimen.

Calculate the RF in IU/ml ( use the following formula )

RF = S x D  

S = Sensitivity of the reagent i.e. 10 IU/ml

D = Highest dilution of serum showing agglutination.
